What Magnesium Glycinate Does

Magnesium glycinate is a chelated form of magnesium bound to glycine, an inhibitory amino acid. This combination offers two distinct advantages: the glycinate chelation provides exceptionally high bioavailability with minimal gastrointestinal side effects (unlike magnesium oxide or citrate, which can cause diarrhea), and the glycine itself has independent calming properties via GABA and glycine receptor modulation. Magnesium participates in over 300 enzymatic reactions including ATP production, DNA synthesis, protein folding, and neuromuscular function.

Why glycinate over other forms? Magnesium oxide has only ~4% bioavailability. Citrate is better (~30%) but causes osmotic diarrhea at higher doses. Glycinate achieves ~80% absorption with virtually no GI distress, and the glycine moiety adds calming and sleep-promoting effects — a dual benefit from a single supplement.

Recent Research (2025–2026)

A November 2025 precision-based randomized placebo-controlled trial (PPCCT) published in The American Journal of Clinical Nutrition found that magnesium glycinate supplementation increased the abundance of gut bacteria (Carnobacterium maltaromaticum and Faecalibacterium prausnitzii) that synthesize vitamin D locally, with this locally produced vitamin D helping to inhibit the development of colorectal cancer cells. A December 2025 RCT in 153 participants with sleep problems found that magnesium bisglycinate improved insomnia symptoms compared to placebo. A 2026 animal study investigating chronic organic magnesium supplementation found that magnesium glycinate exhibited anxiolytic properties, reducing anxiety-like behaviors — though further clinical studies are warranted to validate these effects in humans.
