What Is 7-Keto DHEA

7-Keto DHEA (3-acetyl-7-oxo-dehydroepiandrosterone) is a naturally occurring metabolite of DHEA, the most abundant steroid hormone precursor in the human body. Unlike parent DHEA, 7-Keto cannot be converted into testosterone or estrogen, making it a non-hormonal alternative for people seeking the metabolic benefits of DHEA without the risk of altering sex hormone levels. DHEA production peaks in the mid-20s and declines roughly 2% per year thereafter, and 7-Keto follows the same decline. Supplementation restores thermogenic signaling that naturally diminishes with age.

Mechanism of Action

7-Keto DHEA increases metabolic rate primarily by upregulating three thermogenic enzymes in the liver: fatty acyl-CoA oxidase, malic enzyme, and glycerol-3-phosphate dehydrogenase. These enzymes drive mitochondrial fatty acid oxidation and heat production. The net effect is an increase in resting metabolic rate without stimulant-like side effects — no jitteriness, elevated heart rate, or insomnia.

Additionally, 7-Keto influences thyroid hormone activity. It enhances the peripheral conversion of T4 to the active T3 form, which is a key driver of basal metabolic rate. This is particularly valuable during caloric restriction, when the body down-regulates thyroid output as a survival mechanism. By maintaining T3 levels, 7-Keto helps prevent the metabolic slowdown that commonly stalls fat loss during dieting.

Because the 7-oxo group on the molecule blocks the enzymatic pathways (3-beta-HSD and 17-beta-HSD) that would otherwise convert DHEA into androgens or estrogens, 7-Keto is structurally incapable of affecting sex hormone levels. This has been confirmed in clinical studies showing no changes in testosterone, estradiol, or PSA after supplementation.

Dosing Protocol

Brand: Life Extension — 7-Keto DHEA Metabolite, 100 mg

TimeDoseNotes
4:30 AM (fasted)100 mgFirst dose upon waking, on empty stomach
12:00 PM (midday)100 mgSecond dose at midday for sustained thermogenic effect

Total daily dose: 200 mg. The split dosing maintains elevated thermogenic enzyme activity throughout the active hours. See the full timing context in the Supplement Stack.

Scientific Evidence

Body Composition Improvement

A randomized, double-blind, placebo-controlled trial (Zenk et al., 2007) found that overweight adults supplementing with 200 mg/day of 7-Keto DHEA for 8 weeks lost significantly more body weight and body fat than the placebo group. The 7-Keto group lost 2.88 kg vs. 0.97 kg in the placebo group, with no adverse effects on blood pressure, heart rate, or blood chemistry. PMID: 17297159

Resting Metabolic Rate Increase

Kalman et al. (2000) demonstrated that 7-Keto DHEA supplementation (100 mg twice daily) combined with exercise increased resting metabolic rate (RMR) by 1.4% above baseline, while the placebo group experienced a 3.9% decrease in RMR. The net metabolic advantage was over 5%, representing approximately 115 extra calories burned per day. PMID: 12777575

Lab Tests to Monitor

While 7-Keto DHEA has an excellent safety profile, monitoring these markers is how you check whether you are getting the intended metabolic benefits, and catch any unexpected shifts early.

Practical Takeaways

  • 7-Keto DHEA is a non-hormonal metabolite — it cannot convert to testosterone or estrogen, making it safe for long-term use without sex hormone concerns.
  • Its primary value is preventing the metabolic slowdown that occurs during caloric restriction by maintaining T3 levels and thermogenic enzyme activity.
  • Split dosing (100 mg AM + 100 mg midday) maximizes the thermogenic window across waking hours.
  • Clinical trials show a net 5% metabolic advantage over 8 weeks, translating to roughly 115 extra calories burned daily.
  • Monitor DHEA-S and TSH at baseline and every 6 months to confirm the supplement is working as intended without disrupting the adrenal or thyroid axis.

Recent Research (2025–2026)

A 2023 systematic review published in Current Nutrition Reports analyzed all available 7-Keto DHEA trials and found that half of the studies with low risk of bias demonstrated significant reductions in body weight, body fat percentage, and BMI, while two studies confirmed increases in resting metabolic rate. The review concluded that 7-Keto DHEA's thermogenic mechanism — elevating energy expenditure without affecting sex hormones — remains its primary clinical advantage, though larger and longer trials are still needed. A separate analysis noted that 7-Keto DHEA was 2.5 times more thermogenic than its parent compound DHEA in vitro, reinforcing its role as a calorie-deficit support agent rather than a standalone fat burner.
